Coming from the south, out of, lets say Frankfurt, you travel the A5 "Autobahn" north to the "Gambacher Kreuz" and switch over to the Autobahn "E45" traveling towards "Wetzlar", "Herborn", "Haiger", "Dortmund". You continue on the "A45" until you reach the "Giessener Süd Kreuz" exit following the signs to "Giessen".
